212282 Bacterial Community Structure In a Hydrogen-Based Membrane Biofilm Reactor

Abstract. The whole bacterial community structure in a novel hydrogen-based membrane biofilm reactor (MBfR) was clarified by PCR-DGGE and high-density universal 16S rRNA Microarray, which includes 506,944 probes targeted to 8935 clusters in 16S rRNA gene sequences. The PCR-DGGE results shown bacterial communities evolved with operation time in MBfR. It was obviously that the bacterial number increased as time increased. The Microarray results shown 360 and 997 bacterial subfamilies were detected in MBfR operated at 10 days and 40days. The whole bacteria can separate as 19 and 24 phyla in two samples, respectively. Proteobacteria was the predominant phylum, 270 and 505 subfamilies were found in two samples. Alphaproteobacteria was the dominant classes in Proteobacteria and occupied 39.3% and 45.5% in two samples, respectively. Acidobacteria, Firmicutes and Bacteroidetes were the subdominant groups, occupying around 14.2%, 11.2% and 11.2% in MBfR at stable stage. Some bacteria groups included Cyanobacteria, Verrucomicrobia, Planctomycetes, Chloroflexi and Spirochaetes phylum also found more than 15 subfamilies. All the results shown the MBfR system had more bacterial community diversity in stable stage. Moreover, the phylogenetic trees of top 50 bacteria were constructed. It were selected by compared the relative average fluorescence intensity of the whole bacteria in MBfR.
