[Objective] To study the extraction technology of theabrownine(TB) from Liubao tea,and its liver protection on acute hepatic injury induced by carbon tetrachloride ( CCl4 ) in mice.[Method] Orthogonal test was adopted to investigate effects of solid-liquid ratio,extracting temperature,time,times on yield of Liubao tea theabrownine,the technique was optimized,and the liver protection of theabrownine was analyzed. [Result] The results showed that effects of extraction times and temperature on theabrownine yield reached significant level,while other two factors were not significant. The optimum extraction conditions were determined as follows: solid-liquid ratio 1:30, extraction temperature 100 °C, extraction time 80 min,extraction 4 times. Under this condition, the yield could be up to 22. 92%. Results of liver protection research showed that, following the treatment with TB,alanine aminotransferase( ALT) and aspirate aminotransferase (AST) activities and malondialdehyde (MDA) content were significantly reduced and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities and the glutathione (GSH) content were obviously increased in the mice of treatment group. [Conclusion] Liubao tea has abundant theabrownine, and TB have protective effects on acute hepatic injury induced by CCl4 in mice, which will lay a solid foundation for deeply development of Liubao tea.
