By changing the nucleation mechanism, the phenomenon of oilingout affect the crystallization process of Active Pharmaceutical Ingredients obviously. The nucleation in the oiling out system of erythromycin ethylsuccinate was investigated in this study. Antisolvent crystallization experiments of oilingout system were studied at different supersaturations with the same temperature. For successful control of nucleation events, several process analytical technology (PAT) tools such as Focused Beam Reflectance measurement (FBRM) and partical vision measurement (PVM) have been introduced. The induction time of the oiling-out system were determined and correlated with the nucleation kinetics model. Depending on the twostep nucleation theory, the position of nucleation points is determined by the primary nucleation process. The results also revealed that the supersaturation generated played key roles in the nucleation mechanism. For the different supersaturation generation experiments, the homogeneous nucleation process were observed with a high supersaturation; on the contrary, heterogeneous nucleation taken place with a low supersaturation.
