Lasso peptides are unique ribosomally synthesized natural products that exist in a topologically constrained conformation.  Recently we have uncovered some mechanistic details about the biosynthesis of these peptides in their bacterial hosts.  This talk will discuss the role of the leader peptide in lasso peptide biosynthesis as well as the promiscuity of the maturation enzymes.
