Dynamic Impact Of Water Thermodynamics On Normal Phase Chromatography

David Willcox, Process Development, Abbott Laboratories, 1401 Sheridan Road, Dept R452-R13-2, North Chicago, IL 60091

Water was found to have a significant impact on a normal phase chromatography process. This was studied systematically in order to understand the impact of water on the elution, the thermodynamics of water adsorption in the solvent systems used, and the dynamics of water adsorption / desorption. This information was used to optimize two chromatography processes by dynamically controlling the water throughout the process.
